Is it recommended to shut down all running VMs to upgrade from 4.5.0 to 4.5.1?

No, that is not a requirement or a recommendation. The upgrade should not affect the running VMs or vice-versa.

Most, if not all, of the upgrades I have seen leave the VMs running.
